摘要: The synergism between the hypnotic effects of chloral hydrate and ethanol was investigated in male mice by determination ED5O9s use isobolographic method. A significant potentiation observed for mixtures which these agents were present a weight ratio equal to or greater than 1:7.2, simple additivity being with ratios smaller 1:7.2. Investigation trichloroethanol, an active metabolite hydrate, respectively, revealed no departures from additivity, thus leading conclusion that interaction must be one directly. also characterized observations compositions exhibited shorter duration action equipotent doses administration two minutes prior resulted further potentiation. Synergism toxic 24-hour, nonaggregated LD5O9s. Mixtures 3.6:1 (1:1 molar ratio) potentiation, whereas those 1:5, 1:10 1:15 showed partial but antagonism. No seen 1:1, 1:1.14, 1:3 1:30. Therapeutic standard safety margins hydrate-ethanol 3.6:1, 1:1 1:1.14 all found significantly respective values either ethanol.
