摘要: The cocaine analog[3H]2β-carbomethoxy-3β-(4-fluorophenyl)tropane ([3H]CFT or [3H]WIN 35,428) binds with high affinity and selectivity to receptors in the monkey caudate-putamen. [3H]CFT was used map regional distribution of binding sites slide-mounted sections brains using autoradiographic techniques. Hemicoronal brain were incubated (3 nM) alone presence excess (−)-cocaine (30 μM) mask sites. High densities detecte caudate nucleus, putamen, nucleus accumbens. In all three regions, markedly reduced by coincubatin unlabeled (−)-cocaine, indicating low levels nonspecific binding. Little no observed cortex, thalamus, globus pallidus, white matter tracts at studied. order characterize biding for tissue sections, competition experiments conducted a fixed concentration range concentrations (+)-cocaine, CFT, Lu 19-005, GBR 12909, bupropion, citalopram. IC50 values drugs corresponded closely their reproted caudate-putamen membranes (r =0.99, p <0.0001), suggesting that is similar two preparations. These findings support view labeled are localized predominantly dopmaine-rich regions implicated behavioral effects abuse cocaine.
