Silencing adenosine A2a receptor enhances dendritic cell-based cancer immunotherapy.

摘要: Overexpression of adenosine in the tumor region leads to suppression various immune cells, particularly T cells through ligation with 2a receptor (A2aR). In this study, we intended increase efficacy lysate-loaded DC vaccine by silencing expression A2aR on application A2aR-specific siRNA-loaded PEG-chitosan-lactate (PCL) nanoparticles (NPs) 4T1 breast tumor-bearing mice. Combination therapy and NPs markedly induced regression increased survival time These ameliorative effects were partly via downregulation immunosuppressive function cytotoxic lymphocytes, induction immune-stimulatory cytokines. Moreover, combination could suppress angiogenesis metastasis processes. results imply novel for treatment cancer using which can be translated into initial phase clinical trials near future.
