摘要: For precise determination of the catalytic activity 3-hydroxy-3-methylglutaryl-coenzyme A (HMG-CoA) reductase (EC 1.1.1.34), HMG-CoA employed as substrate must be free HMG, CoA, and other inhibitors activity. The standard purification by paper chromatography gives poor resolution from CoA may accompanied some decomposition HMG-CoA. We describe a simplified procedure for synthesis isolation reaction mixture homogeneous, high specific [3(-14)C]HMG-CoA or nonpolar contaminants. Isolation utilizes ion-exchange in gradient ammonium formate, which is subsequently removed lyophilization. methods are proposed use preparation HMG0CoA.
