摘要: Standard light microscope histological evaluation of peripheral nerve lesions has been used routinely to assess demyelination; however, the development magnetic resonance (MR) methodology for assessing may provide complimentary information, with less expense and in time than histology methods. In this study, utility multicomponent NMR T2 relaxation analysis myelin injury toxicology studies was examined using two dithiocarbamates, N,N-diethyldithiocarbamate (DEDC) pyrrolidine dithiocarbamate (PDTC), known produce elevate copper nervous system. conjunction standard methods determine if dithiocarbamate-mediated accumulation associated more severe lesions. Male Sprague-Dawley rats were administered i.p. DEDC 8 weeks maintained on either a diet containing normal (13 ppm) or elevated (200 copper. Another group male oral PDTC 200 ppm diet, controls given only 47 weeks. Following exposures, morphology sciatic evaluated microscopy excised fixed nerves; levels determined ICP-AES. Light demonstrated presence primary myelinopathy dithiocarbamate-exposed characterized by intramyelinic edema, demyelination, secondary axonal degeneration. Both level number degenerated axons, as ascertained ICP-AES respectively, augmented dietary supplementation administration PDTC. revealed decreased contribution from shortest component spectra obtained animals PDTC, consistent content; decrease water inversely correlated lesion counts. Also, showed reduced variability compared assessment. These support complementary method microscopic evaluations that also be applicable vivo assessments.
