摘要: We report on a pilot study showing proof of concept for the passive delivery nanoshells to an orthotopic tumor where they induce local, confined therapeutic response distinct from that normal brain resulting in photothermal ablation canine transmissible venereal (cTVT) model. cTVT fragments grown severe combined immunodeficient mice were successfully inoculated parietal lobe immunosuppressed, mixed-breed hound dogs. A single dose near-IR (NIR)-absorbing, 150-nm was infused i.v. and allowed time passively accumulate intracranial tumors, which served as proxy metastasis. The accumulated within cTVT, suggesting its neovasculature represented interruption blood-brain barrier. Tumors thermally ablated by percutaneous, optical fiber-delivered, NIR radiation using 3.5-W average, 3-minute laser at 808 nm selectively elevated temperature tissue 65.8 +/- 4.1 degrees C. Identical doses applied white gray matter contralateral side yielded sublethal temperatures 48.6 1.1 designed minimize thermal damage absence compensate variability accumulation tumor. Postmortem histopathology treated sections showed effectiveness selectivity nanoshell-assisted ablation.
