摘要: The PRP17 gene of the yeast Saccharomyces cerevisiae encodes a protein that participates in second step splicing reaction. It was found recently is identical to cell division cycle CDC40 gene. PRP17/CDC40 codes for with several copies WD repeat, motif large family proteins play important roles signal transduction, progression, splicing, transcription, and development. In this report, we describe identification human, nematode, fission homologues PRP17/ S. cerevisiae. newly identified share homology budding throughout their entire sequence, similarity being greatest C-terminal two thirds includes conserved repeats. We show yeast‐human chimera, carrying hPRP17 protein, able complement defects prp17 mutant. Moreover, chimeric co-precipitate intron‐exon 2 lariat intermediate intron product, providing evidence these are spliceosome-associated. These results functional conservation Prp17 evolution suggest takes place by similar mechanism eukaryotes.
