Dispersed pancreatic graft cryopreservation in the dog: In vivo assessment of preservation protocols

摘要: Dispersed canine pancreatic grafts were cryopreserved and the in vivo function was studied following intrasplenic autotransplantation. Four protocols employed, examining effects of cooling thawing rates cryoprotectant (dimethylsulfoxide) concentration on graft survival. The degree injury by each protocol assessed requirement for exogenous insulin transplantation. Cooling at 5 °C/min 80 allowed three successful from seven when thawed using 1.4 or 2 M Me2SO but only one success eight 8 °C/min. Of experiments where preservation achieved estimated as less than 50% four it probably greater 50%. Each exhibited considerable variability islet When sufficient mass transplanted to restore fasting euglycaemia, function, glucose-stimulated release intravenous glucose disposal, identical fresh grafts.
