Chronic clonazepam administration induced benzodiazepine receptor subsensitivity

摘要: Clonazepam and chlordiazepoxide were administered chronically in increasing doses for three weeks two different strains of mice. Forebrain [3H]diazepam binding was assayed groups mice sacrificed at 2, 26, 50 hr 10 days following the last dose. Scatchard single point analyses revealed a significant decrease number sites [Bmax] which persisted least chronic clonazepam treatment. The Bmax changes observed treatment less pronounced than those elicited by clonazepam. No receptor affinity (Kd) detected with either drug. In clonazepam-treated animals, values returned to normal day after drug Chronic benzodiazepine administration therefore induced apparent mouse forebrain. magnitude duration subsensitivity appears depend on potency benzodiazepine.
