摘要: P-glycoprotein is implicated in clinical drug resistance; thus, rapid quantitative analysis of its expression and activity paramout importance to the design success novel therapeutics. The scope for application imaging image tools this field reported here at "proof concept" level. was utilized as a model immunofluorescence subsequent spatial intensity distribution (SpIDA). Following studies, p-glycoprotein inhibition function verapamil concentration assessed two cell lines using live intracellular Calcein retention routine monolayer fluorescence assay. Intercellular sub-cellular distributions transporter between parent MDR1-transfected Madin-Derby Canine Kidney were examined. We have demonstrated that can provide dose-response parameters while permitting direct microscopic fluorophore fixed samples. Analysis with SpIDA offers ability detect heterogeniety labeled species, conjunction staining may be applied determination pharmacological or biopsies providing prognostic tool.
