摘要: Reproducible hordein electrophoregrams were obtained when hordeins, extracted from Canadian-grown barley cultivars, subjected to polyacrylamide gel electrophoresis at acid pH (acid-gel PAGE). The hordeins room temperature into a solution of 55% (vol/vol) isopropanol containing 2% monothioglycerol. Extraction high (60 °C) produced significant reduction in the number protein bands available for cultivar identification. Alkylation reduced prior acid-gel PAGE, did not improve resolution or appearance electrophoregrams. effect environment on electrophoregram was studied. Four cultivars grown eight locations Western Canada during 2 successive years. Hordein qualitatively independent growth location, year and content. These results suggest that PAGE should prove useful as technique
