摘要: — The present experiments were designed to provide direct evidence that the increase in dopamine β-hydroxylase (DBH) activity after treatment with reserpme results from an augmented synthesis of new enzyme protein. After vivo had shown DBH could not be labelled a sufficient extent even administration high doses (8.3 mCi/kg) [3H]leucine we took advantage earlier observations neuronally-mediated induction initiated progresses organ cultures adrenal medullae similar manner as vivo. With system it was possible achieve labelling molecules and produced by reserpine really due specific synthesis. fact half-life identical controls reserpine-treated animals eliminated possibility increased slower rate degradation. Both culture showed very clearly immunoprecipitation has followed additional purification gel electrophoresis since spite use monospecific antibodies careful washing immunoprecipitates contaminating coprecipitated accounted for up 96% total precipitated radioactivity whole adrenals (in experiments) 80% medulla (organ experiments). coprecipitation contaminants most probably addition carrier proteins, used order make precipitation small amounts proteins visible.
