Roles of lipid-modulating enzymes diacylglycerol kinase and cyclooxygenase under pathophysiological conditions

摘要: Lipid not only represents a constituent of the plasma membrane, but also plays pivotal role in intracellular signaling. Lipid-mediated signaling system is strictly regulated by several enzymes, which act at various steps lipid metabolism. Under pathological conditions, prolonged or insufficient activation this results dysregulated signaling, leading to diseases such as cancer metabolic syndrome. Of lipid-modulating diacylglycerol kinase (DGK) and cyclooxygenase (COX) are intimately involved system. DGK consists family enzymes that phosphorylate second messenger (DG) produce phosphatidic acid (PA). Both DG PA known activate molecules protein C. COX catalyzes committed step prostanoid biosynthesis, involves metabolism arachidonic prostaglandins. Previous studies have shown engaged number conditions. This review summarizes functional implications these two ischemia, liver regeneration, vascular events, diabetes, inflammation.
