摘要: Five differently preserved groups of adult Rhipicephalus appendiculatus specimens were compared for quality DNA extracted. Three methods used to extract from i.e. two simple mosquito validated extraction and a tick method. Extraction legs was attempted. The extracted evaluated by the success PCR amplification ITS2 gene mitochondrial COI fragment. Fresh (i.e. killed just before extraction) had highest followed in ethanol subsequently stored refrigerator (4 °C). There no significant difference between cryopreserved 70% specimens. It possible amplify ticks. Sequenced amplicon template obtained ticks as legible those whole extract. showed significantly lower than protocol.
