摘要: Promoting remyelination is crucial for patients with demyelinating diseases including multiple sclerosis. However, it still a circuitous conundrum finding practical remyelinating therapy. Electroacupuncture (EA), originating from traditional Chinese medicine (TCM), has been widely used to treat CNS all over the world, but role of EA in barely known. In this study, we examined properties and mechanisms cuprizone-induced model, murine model By feeding C57BL/6 mice chow containing 0.2% cuprizone 5 weeks, successfully induce demyelination as proved by weight change, beam test, pole histomorphology, Western Blot. treatment significantly improves neurobehavioral performance at week 7 (2 weeks after withdrawing chow). RNA-seq RT-PCR results reveal up-regulated expression myelin-related genes, myelin associated protein (MBP, CNPase, O4) are also increased treatment, indicating therapeutic effect on model. It acknowledged that microglia exert phagocytic degraded debris clear these detrimental debris, which necessary process subsequent remyelination. We found enhanced clearance detected dMBP staining red oil O staining. Our further studies suggest more assemble area (corpus callosum) during cells inside corpus callosum mostly plump, ameboid, shape, quite different ramified outside callosum. result unravels most genes relating positive regulation phagocytosis (GO:0050766) up-regulated, treatment. During clearance, tend change their phenotype toward M2 phenotype. Thus, probed into our study. Immuno-staining show CD206 Arg1, ratio CD206/CD16/32 higher group. conclusion, demonstrate first time enhances removal activated demyelination, promotes
