摘要: Abstract Allelic loss studies have suggested that a glioma tumor suppressor gene resides in 425-kb region of chromosome 19q, telomeric to D19S219 and centromeric D19S112. Exon amplification cosmid contig spanning this yielded four exons with high homology rat protein serine-threonine phosphatase from approximately 100 kb D19S219. Isolation near full-length cDNA human fetal brain library revealed tetratricopeptide motif, almost identical PPP5C (PP5) highly homologous PPT. Northern blotting demonstrated expression most tissues, including brain. Primary cultured gliomas were studied for genetic alterations using pulsed-field gel electrophoresis, routine Southern blots, genomic DNA- RNA-based single-strand conformation polymorphism analysis. Genomic not detected any the gliomas, all expressed gene, suggesting is putative 19q gene.
