摘要: An oxygen microsensor was used to measure internal profiles in biocatalyst particles of different diameter and activity. The were made agarose gel contained an reducing enzyme, L-lactate mono-oxygenase. kinetics the enzyme could be well described by Michaelis-Menten equation. From substrate concentration profile intrinsic kinetic parameters determined means fitting a simulated measurements, using Marquardt's algorithm. found following this procedure appeared independent particle radius or loading used, proving method reliable. These also compared with free which biological monitoring system. showed decrease factor 2.3 for V(m) value 2.7 K(m) enzyme. conclusion can drawn that immobilization as such carrier material not only have effect on maximum conversion rate (V(m)) but affinity (K(m)) oxygen.
