摘要: Abstract Background The use of non-cultured autologous cells to promote wound healing and in reconstructive procedures is increasing. One common method for preparing these the ReCell ® device. However, despite its current clinical use, no characterisation cell suspension produced using a device has been published. Objective To characterise that applied wounds type, viability, yield, stability proliferative potential. Methods was used harvest from 2 cm 2 piece split-thickness skin isolated dermatome. resulting analysed viability over time, potential reproducibility. Results Average viable yield 1.7 × 10 6 /cm tissue, with 75.5% total isolate viable. Total number not significantly reduced after 4 h storage at 22 °C or 4 °C, stable 24 h 4 °C. Proliferative assessed colony forming assay, 0.3% keratinocyte colonies. Predominantly contained keratinocytes (64.3 ± 28.8%) fibroblasts (30.3 ± 14.0%), small population melanocytes also identified (3.5 ± 0.5%). Finally, supernatant low protein (0.92 mg/ml) had significant effects on growth when ex vivo . Conclusions These results suggest provides preparation high potential, containing apparent toxic debris. Further work sustained , particular application wound, will be important better understand clinic.
