Identification and Phenotype of MAIT Cells in Cattle and Their Response to Bacterial Infections.

摘要: Mucosal-associated invariant T (MAIT) cells are a population of innate-like that utilize semi-invariant cell receptor (TCR) α chain and restricted by the highly conserved antigen presenting molecule MR1. MR1 presents microbial riboflavin biosynthesis derived metabolites produced bacteria fungi. Consistent with their ability to sense ligands from bacterial sources, MAIT have been associated immune response variety infections, such as Mycobacterium spp., Salmonella spp. Escherichia coli. To date, studied in humans, non-human primates mice. However, they only putatively identified cattle PCR based methods; no phenotypic or functional analyses performed. Here, we utilizing tetramers high-throughput TCR sequencing. Phenotypic analysis revealed features analogous those humans mice, including expression an orthologous TRAV1-TRAJ33 chain, effector memory phenotype irrespective tissue localization, transcription factors PLZF EOMES. We determined frequency peripheral blood multiple tissues, finding enriched mucosal tissues well mesenteric lymph node. Cattle were responsive stimulation 5-OP-RU competent vitro. Furthermore, milk increased cows mastitis. Following challenge virulent bovis, causative agent bovine tuberculosis zoonosis, expressed higher levels perforin. Thus, implicated two major infections cattle. These data suggest functionally excellent large animal model study role important zoonotic infections.
