Analysis of T cell antigen receptor (TCR) expression by human peripheral blood CD4-8- alpha/beta T cells demonstrates preferential use of several V beta genes and an invariant TCR alpha chain.

摘要: CD4-CD8- (double negative [DN]) alpha/beta T cells are a largely uncharacterized subpopulation of unknown function. To investigate whether these selected to recognize particular antigens or antigen-presenting molecules, DN were purified from the peripheral blood five normal donors and their cell receptor (TCR) alpha beta chains examined. Random cloning TCR by single-sided polymerase chain reaction (PCR) amplification identified an invariant rearrangement between V 24 J Q, with no N region diversity, which was expressed preferentially all donors. also precise 7.2-J (IGRJa14) rearrangement, two variable amino acids encoded in V-J junction, enriched preparations some, but not all, Analysis quantitative PCR demonstrated that expression four gene families, 2, 8, 11, 13, markedly increased preparations. The TCRs multiple indicates cells, at least this phenotype, limited spectrum suggests they may use nonpolymorphic molecules.
