Rapid detection of potato late blight using a loop-mediated isothermal amplification assay

摘要: Abstract Potato late blight caused by Phytophthora infestans is one of the most destructive plant diseases that threaten global food security. Early and effective diagnosis P. required before disease management decisions are made. Here, we developed a quick protocol to detect based on loop-mediated isothermal amplification (LAMP) assay. The P . i nfestans s pecific m ultiple c opy DNA sequences (PiSMC), transposon-like element, provides an ideal target for molecular detection this pathogen. We designed highly specific sensitive primers allowing LAMP pathogen at 64°C in 70 min. In validation assay, all 15 isolates collected from China, Europe South America could be positively detected, but results other 9 species infecting different plants, fungal bacterial pathogens tested were negative. limit assay 1 pg DNA. Moreover, LAMP-PiSMC able infected leaves, tubers soil. Taken together, study reports development early potato blight.
