摘要: Summary The ESCRT-I and ESCRT-II supercomplex induces membrane buds that invaginate into the lumen of endosomes, a process central to lysosomal degradation ubiquitinated proteins. solution conformation membrane-budding ESCRT-I-II from yeast was refined against small-angle X-ray scattering (SAXS), single-molecule Forster resonance energy transfer (smFRET), double electron-electron (DEER) spectra. These refinements yielded an ensemble 18 ESCRT-I-II structures range compact highly extended. crescent shapes provide basis for detailed mechanistic model, in which stabilizes coordinates cargo sorting by lining pore nascent bud necks. hybrid refinement used here is general should be applicable other dynamic multiprotein assmeblies.
