Production of Acrylamide using Alginate‐Immobilized E. coli Expressing Comamonas testosteroni 5‐MGAM‐4D Nitrile Hydratase

摘要: A thermally-stable nitrile hydratase produced by Comamonas testosteroni 5-MGAM-4D has been expressed in Escherichia coli, and the alginate-immobilized transformant evaluated as catalyst for conversion of acrylonitrile to acrylamide. In batch reactions with recycle, productivity decreased increasing concentration or reaction temperature, but was relatively insensitive acrylamide concentration. total 206 consecutive recycle were run at 5 °C 1035 g acrylamide/g dry cell weight 95 g acrylamide/L; initial final volumetric productivities this series 197 96 g acrylamide/L/h, respectively. packed column reactor yielded lower than reactions, presumably due contact fixed-bed a constant high acrylonitrile.
