Simple and Noninvasive Breath Test Using 13C-Acetic Acid to Evaluate Gastric Emptying in Conscious Rats and Its Validation by Metoclopramide

摘要: Abstract The 13C-breath test has been used to clinically evaluate gastric emptying. However, this method not sufficiently validated in experimental animals. present study aimed establish a simple and noninvasive 13C-breath-test system Sprague-Dawley male rats. After fasting, rats were orally administered Racol containing 13C-acetic acid housed desiccator. expired air the chamber was collected breath-sampling bag using tube aspiration pump. level of 13CO2 measured an infrared spectrometer at appropriate intervals for 120 min. During period, rate excretion increased, peaked, decreased thereafter. maximum concentration (Cmax) area under curve (AUC120 min) increased volume- dose-dependent manners. time taken reach (Tmax) as volume but affected by dose 13C-labeled acetic acid. Metoclopramide dose-dependently Cmax shortened Tmax compared with those control rats, whereas AUC120 min affected. These results confirm that can successfully Moreover, is suitable investigating additional physiological functions other labeled compounds.
