摘要: The purpose of the study was to investigate impact rat cytomegalovirus (RCMV) infection on development nervous system in embryos, and evaluate involvement Wnt signaling pathway key molecules downstream gene neurogenin 1 (Ngn1) RCMV infected neural stem cells (NSCs). Infection control groups were established, each containing 20 pregnant Wistar rats. Rats group inoculated with by intraperitoneal injection first day pregnancy. Rat E20 embryos taken teratogenic rate. NSCs isolated from E13 maintained vitro. We found: 1) Poor fetal found low survival high malformation rates. 2) proliferation differentiation affected. In group, proliferated more slowly had a lower neurosphere formation rate than control. ratio neurons glial significantly different that control, showed immunofluorescence staining. 3) Ngn1 mRNA expression nuclear β-catenin protein level 2 when differentiated. 4) Morris water maze test performed 4-week pups, rats worse learning memory ability. summary, caused abnormalities embryonic system, inhibited NSC differentiation, Wnt/β-catenin so as affect differentiation. This may be an important mechanism which causes abnormalities.
