Activating transcription factor 3 gene expression suggests that tissue stress plays a role in leiomyoma development

摘要: Objective To determine whether expression of the stress response gene ATF3 and related members activator protein complex-1, cJun cFos, were altered in leiomyoma compared with myometrium, this difference might correlate size or race. Design Laboratory study. Setting University hospital. Patient(s) Fifteen women undergoing hysterectomy for symptomatic leiomyoma. Intervention(s) Tissue procurement, RNA isolation, reverse-transcriptase polymerase chain reaction, real-time (RT-PCR) immunohistochemistry, Western blot. Main Outcome Measure(s) Expression mRNA patient-matched myometrium. Result(s) transcripts decreased matched myometrium by both RT-PCR RT-PCR. The decrease was greater than fivefold a majority samples. reduction seen did not show correlation race size. Surprisingly, immunohistochemistry blot analysis demonstrated an elevation mean 2.9-fold. Transcripts AP-1 genes, significantly −29.57 −23.78 but there no significant change two transcription factors. Conclusions Alterations resemble to mechanical ischemic reported other tissues. Results suggested that increased leiomyoma, may reflect tissue stress.
