摘要: Objective To investigate the levels of matrix metalloproteinases (MMPs), myeloperoxidase (MPO), and tissue inhibitor metalloproteinase-1 (TIMP-1) in tears patients with Stevens-Johnson syndrome (SJS) ocular cicatricial pemphigoid (OCP). Design Prospective, noninterventional cohort study. Participants Four SJS (7 eyes), 19 OCP (37 20 healthy controls who underwent phacoemulsification (40 eyes). Methods Tear washes were collected from all analyzed for MMP-2, MMP-3, MMP-7, MMP-8, MMP-9, MMP-12, MPO, TIMP-1 using multianalyte bead-based enzyme-linked immunosorbent assays. Total MMP activity was determined a fluorometric assay. Correlation studies performed between various analytes within study groups. Main Outcome Measures Levels (in nanograms per microgram protein) total relative fluorescent units minute tears; MMP-8–to–TIMP-1 ratio; MMP-9–to–TIMP-1 correlations MMP-8 MMP-9 both MPO. Results MPO elevated significantly (SJS>OCP) when compared controls. The highest patients, whereas showed lower similar activities. decreased those controls, reaching significance. ratios markedly Across groups, correlated strongly levels, but it did not reach significance patients. There no relationship MMP-7 Conclusions Because are produced by inflammatory cells, particularly neutrophils, correlation data indicate that they may be common source enzymes, including tears. Elevated MMP-to-TIMP suggest an imbalance tear regulation explain predisposition these to demonstrate corneal melting chronic complications associated persistent inflammation.
