Transcriptional regulation of α-fetoprotein expression by dexamethasone in human hepatoma cells

摘要: Abstract The level of alpha-fetoprotein (AFP) mRNA in HuH-7 human hepatoma cells is elevated by the addition dexamethasone to culture medium. To locate DNA region involved hormonal regulation AFP gene, we constructed recombinant plasmids which various lengths 5'-flanking sequence gene were fused CAT gene. Various cell lines transfected with plasmids, incubated or without 3 x 10(-6) M dexamethasone, and then assayed for chloramphenicol acetyltransferase expression. In that produce AFP, treatment resulted stimulated expression when contained 169 base pairs (bp) longer sequence. No effect was observed less than 98 bp long. stimulation effectively suppressed glucocorticoid antagonist RU486, indicating this mediated receptors. 71-bp between positions -169 -98 contains a nucleotide stretch similar consensus responsive element (GRE). Partial alterations decreased response. GRE-containing heterologous (SV40) promoter activity response an orientation- position-independent manner. GRE upstream enhancer function independently from each other.
