摘要: We used a next-generation high-throughput sequencing platform to resequence the Xinguowei and Shouxing melon cultivars, parents of Fengwei melon. found 84% reads (under coverage rate "13×") placed on reference genome DHL92. There were 2,550,000 single-nucleotide polymorphisms 140,000 structural variations in two genomes. also identified 1,290 polymorphic genes between Shouxing. combined specific length amplified fragment (SLAF-seq) bulked-segregant analysis (super-BSA) analyze F2 extreme phenotypes. This method yielded 12,438,270 reads, 46,087 SLAF tags, 4,480 markers (average depth 161.81×). six sweet trait-related regions containing 13 differential markers, 23 sour 48 markers. further fine-mapped trait genomic chromosomes 6, 10, 11, 12. Correspondingly, we mapped 2, 3, 4, 5, 9, Finally, positioned nine 61 candidate parental genome. These corresponded one eight genes. Our study provides basis for marker-assisted breeding desirable traits melons.
