Embryonic lethal mutation in mice induced by retrovirus insertion into the alpha 1(I) collagen gene.

摘要: Experimental insertion of a retrovirus into the germ line mice has resulted in an embryonic recessive lethal mutation. Integration proviral genome occurred at 5' end alpha 1(I) collagen gene, leading to complete transcriptional block. Developmental arrest embryos homozygous mutated allele coincides with high expression gene normal embryogenesis. Insertion mutagenesis by retroviruses may offer general approach identification and isolation genes which are transcriptionally active during mammalian development.
