摘要: Many cell-based regenerative medicine strategies toward tissue-engineered constructs are currently being explored. Cell-cell interactions and with different biomaterials extensively investigated, whereas very few studies address how cultured cells will interact soluble wound-healing mediators that present within the wound bed after transplantation. The aim of this study was to determine adipose tissue-derived mesenchymal stem (ASC), dermal fibroblasts, keratinocytes react when they come in contact deep cutaneous burn bed. Burn exudates isolated from wounds were found contain many cytokines, including chemokines growth factors related inflammation healing. Seventeen identified by ELISA (concentration range 0.0006-9 ng/mg total protein), skin-specific chemokine CCL27. activated both ASC but not keratinocytes, increase secretion CXCL1, CXCL8, CCL2, CCL20. Notably, fibroblasts or showed significant increased vascular endothelial factor (5-fold) interleukin-6 (253-fold), although incorporated bi-layered skin substitute (SS) these differences less pronounced. A similar discrepancy between fibroblast mono-cultures observed recombinant human-CCL27 used instead exudates. Although CCL27 did stimulate any cells, contrast proliferation migration. Taken together, results indicate on transplantation, primarily promote closure. In contrast, and, particular, respond vigorously bed, leading angiogenesis/granulation tissue formation factors. Our findings have implications for choice cell type (ASC fibroblast) be importance taking into account developing advanced therapies difficult-to-close wounds.
