摘要: Angiogenesis is a significant prognostic factor in breast cancer, but the factors that control angiogenesis vivo are not well defined. Multiple angiogenic polypeptides known, and we have determined expression of seven these primary human cancers; relationship to estrogen receptor vascular density was also examined. Vascular endothelial growth (VEGF) its four isoforms (121, 165, 189, 206 amino acids), transforming (TGF)-beta1, pleiotrophin, acidic basic fibroblast (FGF), placental factor, thymidine phosphorylase (platelet-derived cell factor) were quantitated by RNase protection analysis. beta-FGF measured ELISA. The (ER), epidermal receptor, analyzed 64 cancers. All tumors expressed at least six different factors. VEGF most abundant, transcript for 121-amino acid form predominated. Other high levels TGF-beta1. Expression did correlate with ER or density. However, did, correlation coefficient 0.3 (P = 0.03). There associations pleiotrophin FGF 0.001) TGF-beta platelet-derived 0.001). Thus, may involve coordinate regulation some High correlated poor prognosis univariate analysis 0.03), as expression. Basic assessed ELISA more highly than normal tissues (median, 346 microg/ml cytosol; range, 54-1323 versus median, 149; 32-509; P 0.01). Implications therapy broad spectrum agents block features common be useful (e.g., antagonism heparin-binding activity agents), because so many expressed. Inhibiting migration directly toxic endothelium would value combined approach therapy.
