Second Cysteine-rich Region of Epidermal Growth Factor Receptor Contains Targeting Information for Caveolae/Rafts

摘要: Previous studies have shown that ∼60% of the epidermal growth factor receptors (EGFRs) in quiescent fibroblasts are concentrated caveolae/raft fraction from purified plasma membranes. This high degree localization suggests EGFR contains targeting information for lipid domains. We used mutagenesis to determine region receptor controls caveolae/rafts resides juxtamembrane, second cysteine-rich region. A 60-amino acid-long sequence within this is continuous with transmembrane domain was sufficient target and cytoplasmic tails both low density lipoprotein caveolae/rafts. TwoN-linked sugars segment were not required proper targeting, although unglycosylated wild-type did localize properly. conclude that, contrast signals coated pit tail, on extracellular side very close membrane.
