A Hyperresponsive HPA Axis May Confer Resilience Against Persistent Paclitaxel-Induced Mechanical Hypersensitivity.

摘要: Paclitaxel (PAC) treatment is associated with persistent, debilitating neuropathic pain that affects the hands and feet. Female sex biological stress responsivity are risk factors for persistent pain, but it unclear whether these important biologically based confer PAC-induced pain. To determine relative contributions of hypothalamic-pituitary-adrenal (HPA)-axis to mechanical hypersensitivity, we employed a PAC protocol consisting three, 2-week cycles every-other-day doses 1 mg/kg versus saline (Week 1) recovery 2), totaling 42 days, in mature male female Fischer 344, Lewis, Sprague Dawley (SD) rats, known differ HPA axis responsivity. Mechanical sensitivity was operationalized using von Frey filaments, per up-down method. Among PAC-injected SD rats exhibited significantly greater hypersensitivity accumulative compared 344 rats. Lewis were not different from or At end protocol, profound whereas appeared relatively resilient long-term effects statistically their saline-injected counterparts. Sex differences mixed noted only early protocol. Moderate may additional painful PAC. If findings hold humans, clinicians be better able identify persons who at increased risks developing during therapy.
