Conjugated linoleic acid inhibits glucose metabolism, leptin and adiponectin secretion in primary cultured rat adipocytes

摘要: Conjugated linoleic acid (CLA) supplementation has been reported to induce insulin resistance in animals and humans, however, the underlying mechanisms remain unclear. The aim of this study was examine direct effects CLA on leptin adiponectin secretion, two hormones with actions known influence sensitivity. Isolated rat adipocytes were incubated (1-200microM) absence presence (1.6nM). inhibited both basal insulin-stimulated gene expression secretion (-30 -40%, P<0.05-0.01). also production (-20 P<0.05-0.01), but not insulin. (50-200muM) decreased glucose uptake (P<0.05-0.01) significantly increased proportion metabolized lactate (P<0.01). Insulin treatment partially prevented inhibitory induced a significant increase percentage lactate. A strong inverse relationship observed between anaerobic utilization decreases secretion. In addition, lipolysis adipogenic transcription factor PPARgamma by CLA.
