摘要: BACKGROUND: Mesenchymal stem cells (MSCs) have been considered for human regenerative therapy applications, and safe culture expansion protocols are needed especially in the context of interspecies contamination. Human platelet lysate (PL) has proposed as animal serum substitute during vitro MSC expansion. In this work, a simplified efficient method to obtain autologous PL replace cell applications is described. STUDY DESIGN AND METHODS: obtained by freezing centrifugation procedures was tested medium supplement adipose mesenchymal (hASC) culture. Differential proliferation, immunophenotypic changes, differentiation under or fetal bovine (FBS) were assessed. RESULTS: contrast 10% FBS supplementation, population doubling time significantly lower when hASCs cultured with same concentration (PL 22.9 ± 1.5 hr vs. 106.7 ± 6.5 hr, t test, p < 0.05). Furthermore, maintained 2.5% supplementation also showed satisfactory results. Immunophenotypic analysis revealed no differences between cultivated both cultures retained potential differentiate into cells. These results demonstrate that from donor can be used hASC culture. CONCLUSIONS: Taken together, evidence provided platelets single sufficient propagation clinical-scale mitigating untoward side effects associated use animal-derived reagents.
