摘要: The functional domain structure of human DNA topoisomerase IIalpha and Saccharomyces cerevisiae II was studied by investigating the abilities insertion deletion mutant enzymes to support mitotic growth catalyze transitions in topology vitro. Alignment S. sequences defined 13 conserved regions separated less or differently spaced sequences. spatial tolerance spacer addressed linkers. importance assessed through individual domains. We found that exact spacing between most domains is noncritical, as insertions were tolerated with no influence on complementation ability. All domains, however, are essential for sustained enzymatic activity A series carboxy-terminal truncations investigated respect ability viability, cellular localization, properties. analysis showed divergent region dispensable catalytic but contains elements specifically locate protein nucleus.
