摘要: The binding of mammalian MCM complexes to chromatin is cell cycle-regulated and under CDC2 kinase negative control. Here, we investigated the properties CDC6 protein, a candidate regulator MCM. levels were relatively constant during HeLa cycle. In asynchronous cells, was mainly detected in nuclei with immunostaining, but some not extractable nonionic detergent. contrast chromatin-bound MCM, this fraction resistant DNase I treatment, suggesting that it binds detergent- nuclease-resistant nuclear structure. S phase became detectable cytoplasm immunostaining; however, level bound unchanged. G2/M still maintained, which hyperphosphorylated by kinase. These data suggest protein associated specific structure throughout cycle major sites on differ between CDC6. However, co-immunoprecipitation assays chemical cross-linking indicated small part present close
