摘要: Efficient plant regeneration was obtained from a cryopreserved embryogenic cell suspension of sugarcane established leaf derived callus. Pregrowing the cells for three days in MS basal medium supplemented with 0.33 M sorbitol essential to process. The were cooled at rate 0.5°C/min -40°C and then stored liquid nitrogen. Thawing carried out rapidly water +40°C, plated without washing onto filter paper discs placed on semi-solid (MS + 3% sucrose 0.13 mg/1 2,4-D +0.25 BAP 0.25 kinetin zeatin). discs, along cells, transferred same, fresh after five hours. After 24 hours scraped off, incubated 28°C dark two weeks before transfer light. A efficiency 92% (regenerated plants, expressed as percent unfrozen control). Plants regenerated grown maturity greenhouse, morphologically identical control plants.
