摘要: The dynamics of branchial Na+,K+,2Cl− cotransporter (NKCC) and Na+,K+-ATPase (NKA) expression were investigated in brown trout Atlantic salmon during salinity shifts the parr-smolt transformation, respectively. In trout, Western blotting revealed that NKCC NKA abundance increased gradually parallel (30- ten-fold, respectively) after transfer to seawater (SW). hydrolytic activity ten-fold SW-transfer. Following back-transfer fresh water (FW), levels both proteins decreased. immunostaining gill SW-acclimated was strong, mainly localized large cells filament around bases lamellae. FW-acclimated less intense more diffuse. Partial cDNAs secretory NKCC1 isoform cloned sequenced from gills. Two differently sized transcripts detected by Northern but not other osmoregulatory tissues (kidney, pyloric caeca, intestine). these associated protein four- 30-fold, respectively, transformation. α-subunit also transformation though a lesser extent than enzymatic (2.5- eight-fold, respectively). separate series vitro experiments, cortisol directly stimulated mRNA tissue salmonids. study demonstrates coordinated regulation © 2002 Wiley-Liss, Inc.
