Na+/Ca2+ exchange inhibitors modulate thapsigargin-induced Ca2+ and Na+ influx in human lymphocytes

摘要: Thapsigargin has been shown the elevate intracellular Na+ concentration in human lymphocytes, but mechanisms underlying thapsigargin-induced entry are little understood. In present study we investigated changes cytosolic free and Ca2+ lymphocytes after inhibition of Na+/Ca2+ exchange with two structurally unrelated compounds, dimethylthiourea ad bepridil. The increase induced by 5 µM thapsigargin was significantly enhanced presence mM or 40 contrast, both compounds decreased elevation. No effect bepridil on influx observed absence extracellular Na+.
