Crosstalk between kinases and Nedd4 family ubiquitin ligases

摘要: A dazzling array of human biological processes achieves coordination and balance through the posttranslational modification protein residues with phosphate (95 Da) or ubiquitin (8565 Da). Over past years, a reciprocal communication has become recognized between phosphorylating (kinases) ubiquitinating (E3 ligases) enzymes. Such crosstalk occurs when kinase acts on ligase vice versa to modify catalytic activity, substrate specificity, subcellular localization modified enzyme. In this review, we focus nine members Nedd4 family E3 ligases signal transducers such as cell surface receptors, cytosolic kinases, phosphatases, transcription factors. Since kinases are well explored established therapeutic targets, hypothesize that mapping onto signalling networks will provide clues full potential pharmacologically targeting ligases.
