摘要: Purpose To investigate the safety and efficacy of subretinal injection human Wharton’s Jelly-derived mesenchymal stem cells (hWJ-MSCs) on retinal structure function in Royal College Surgeons (RCS) rats. Methods RCS rats were divided into 2 groups: hWJ-MSCs treated group (n = 8) placebo control 8). In treatment group, from healthy donors injected space one eye each rat at day 21. Control received saline same volume. Additional 3 animals with nanogold-labelled for vivo tracking localisation using a micro-computed tomography (microCT). Retinal was assessed by electroretinography (ERG) days before repeated 15, 30 70 after injection. Eyes collected histology, cellular molecular studies. Results No tumor formation detected histology during study period. MicroCT scans showed that stayed localised no systemic migration. Transmission electron microscopy located within space. Histology preservation outer nuclear layer (ONL) but not group. However, there significant differences ERG responses between groups. Confocal evidence expressing markers photoreceptor, Muller bipolar cells. Conclusions Subretinal delay loss ONL RCS rats. appears to be safe has potential differentiate retinal-like cells. The this cell-based therapy dystrophies warrants further studies.
