A micro-method for the study of in vitro lymphocyte transformation to specific antigens

摘要: Abstract A quantitative method for studying blast-transformation of cultured lymphocytes in the presence specific antigens is described. The technique performed 0.3 ml microtrays, using 2.5 × 105 per culture, with an optimal culture period 6 days. Culture are pulsed 4 h [3H]-thymidine, giving maximum incorporation and minimum cellular damage. study also describes some disadvantages lymphocyte responses to phytohaemagglutinin (PHA), as a marker T-cell function. Experiments establish ideal conditions each stage can be adjusted accomodate different antigens, cell types animal well human lymphocytes.
