FOXO transcription factors directly activate bim gene expression and promote apoptosis in sympathetic neurons

摘要: Developing sympathetic neurons die by apoptosis when deprived of NGF. BIM, a BH3-only member the BCL-2 family, is induced after NGF withdrawal in these cells and contributes to withdrawal-induced death. Here, we have investigated involvement Forkhead box, class O (FOXO) subfamily transcription factors regulation BIM expression We find that overexpression FOXO induces promotes death BIM-dependent manner. In addition, FKHRL1 (FOXO3a) directly activates bim promoter via two conserved binding sites mutation abolishes activation withdrawal. Finally, show activity deprivation-induced neurons.
