Evaluation of Genetic Homogeneity in Tissue Culture Regenerates of Jatropha curcas L. using Flow Cytometer and DNA-based Molecular Markers

摘要: The present investigation aimed to evaluate the reliability of in vitro propagation methods for elite genotypes Jatropha curcas L., that maintain genetic integrity tissue culture (TC) regenerates among two regeneration systems developed through direct shoot bud using nodal/apical segments (protocol-A) and vitro-derived leaves (protocol-B) as explants. Random amplified polymorphic DNA (RAPD), intersimple sequence repeat (ISSR), simple (SSR) molecular markers, flow cytometery (FCM) were employed homogeneity TC-regenerates at different passages subcultures. RAPD markers showed fifth-generation both protocols. ISSR stability leaf 10th generation. FCM analysis generation protocol-B 20th protocols, ploidy level. SSR assessment protocols confirmed homogeneity. results demonstrated so far explants origins, large-scale multiplication Jatropha.
