Monocyte-macrophage differentiation in vitro : Modulation by extracellular matrix protein substratum

摘要: The influence of extracellular matrix (ECM) on monocyte-macrophage (mo-mφ) differentiation was investigated using an in vitro model with human peripheral blood mononuclear cells (PBMC) maintained different protein substrata. Macrophage specific markers associated studied were, (a) endocytosis modified proteins; (b) appearance mφ metalloproteinases (MMPs); (c) activities myeloperoxidase (MPO) and β-D-glucuronidase; (d) changes the expression cell surface antigens. As duration monocytes culture increased, a progressive increase rate seen as evidenced by functions such 125[I] acetyl BSA gelatinases A B. Significantly higher production MMPs were found fibronectin (FN) COL I than IV (FN > IV) indicating that contact stromal components differentiate at faster rate. FACS analysis done for phenotypic profile characteristic to mo-mφ showed downregulation CD14 occurring substratum dependent manner viz, I) upregulation CD71 high IV. Intracellular enzymatic MPO significantly decreased irrespective substrata, while β-D-glucuronidase activity increased IV). Pretreatment genistein secretion MMPs, particularly MMP 9 ECM phosphorylation signalling process mediating effect. results these studies suggest apart from diffusible factors, microenvironment provided various proteins FN can modulate differentiation.
