摘要: Paraffin-embedded tissues are often the only available material to perform polymerase chain reaction (PCR)-based analysis in various medical purposes. Unfortunately, use many countries of acid fixatives such as Bouin's fluid limits a for molecular analysis. This article reports methodological details DNA purification technique from Bouin-fixed and paraffin-embedded samples based on double washing, an alcohol then aqueous medium, DNA, which enables PCR reactions this material. Comparison results with those obtained by organic solvent frozen tissue fragments showed excellent reproducibility terms detection amplification product agarose gel. However, differences between methods were quite frequently seen allelic typing profile microsatellite sequences (CA repeats), either neo-alleles or loss normal alleles fixed materials that constitute limitation using substrate fine allelotyping.
