Long-term continuous sildenafil treatment ameliorates corporal veno-occlusive dysfunction (CVOD) induced by cavernosal nerve resection in rats.

摘要: It was recently reported in the rat that vardenafil given a continuous long-term manner successful preventing smooth muscle fibrosis penile corpora cavernosa and corporal veno-occlusive dysfunction (CVOD) occur following bilateral cavernosal nerve resection (BCNR), model for human erectile after radical prostatectomy. To expand on this finding to determine whether effect common other PDE5 inhibitors, occurred part by stimulation of spontaneous induction inducible nitric oxide synthase (iNOS, also known as NOS2), male Fischer 344 rats (N=10/group) were subjected either BCNR or unilateral (UCNR) treated with sildenafil (20 mg kg(-1) day(-1)) drinking water daily 45 days. Additional groups received L-NIL (6.7 inhibitor iNOS activity, without concurrent administration. determined sildenafil, like vardenafil, (1) prevented 30% decrease cell/collagen ratio, 3-4-fold increase apoptosis reduction cell proliferation, partially counteracted collagen, seen both UCNR BCNR; (2) normalized CVOD, measured dynamic infusion cavernosometry, induced UCNR. The inhibition activity exacerbated CVOD rats, but functional effects not affected L-NIL. These data suggest salutary inhibitors function post-cavernosal involve their ability prevent alterations histology damage, process apparently independent from endogenous induction.
