Calcium incorporation in cultured chondroblasts perturbed by an electromagnetic field

摘要: We tested the hypothesis that electric perturbation influences 45Ca incorporation in extracellular matrix (ECM) of cartilage vitro. Hypertrophic chondroblasts tibial epiphyses (HC), sternum (SC), and skin fibroblasts (F) were cultured from chick embryos. HC, SC, F cells micromass seeded three times per week maintained at 37.5 degrees C with 5% CO2 for two weeks. Cultures randomly designated control (C) or exposed (E) to a pulsed electromagnetic field (PEMF). A time course experiment calcium all groups showed 24 h exposure produced largest biological response chondroblasts. Calcium required supplemental phosphate. Autoradiography data indicated into macromolecules largely occurred ECM. steady-state was enhanced by Streptomyces hyaluronidase (SH) but not testicular (TH). experiments effects phosphate, SH, TH, PEMF alone various combinations on these cultures. Only SH plus phosphate incorporation. Other examined effect rotenone freeze-thawing PEMF. freeze-thaw HC only. appeared cause disruption ECM, enhancing probability calcification.
